Tuberculosis is a global emergency particularly in developing countries. AndhraPradesh is the third largest state in India. Ourdata based on 3379 subjects. Amongwhich 55.8% are new sputum positive cases and males were more affected than female. Theage group from35-44 and 45-54 are more proneTB bacteria.Young children below14 years are less susceptible. We did not found any TB patients with HIV infection. Thepopulation attributable risk is 29.89%

A New Facultative Alkaliphilic, Potassium Solubilizing, Bacillus Sp . S VUNM 9 Isolated f rom Mica Cores of Nellore District, Andhra Pradesh, India

A new facultative alkaliphilic Bacillus speciesisolated from mica mines of Nellore district of Andhra Pradesh, India. This strain is gram positive, rod shaped, motile bacteria capable of growth in aerobic conditions up to pH 12.0 exhibiting optimum growth at pH 10.0. The strain is oxidative, catalase positive, urease positive, able to hydrolyse starch, gelatine, able to utilize lactose, D-glucose and sucrose and produce acids. It also converts nitrate into nitrite. Further, exhibited positive results for methyl red and Voges Proskauer reaction. The strain showed optimum growth at 0.5% NaCl and able to thrive even at higher concentration of sucrose. The 16S rDNA sequence showed 99% similarity with Bacillussp19(HQ433576) and closest relative is Bacillus amyloliquifaciens (JN086147).The efficiency of this strain for potassium dissolution from insoluble mica was evaluated in vitro. Modified Alexandrov’s medium supplemented with 0.1% mica powder was used to investigate the potassium solubilising activity of the Bacillus spSVUNM9strain. Final pH, total acidity, soluble K content and released organic acids were determined in culture media after 21days of incubation. The K content was increased to 2.5times greater than the control. Mineral potassium solubilisation was directly related to the pH drop by the strain. The analysis of the culture medium by high pressure liquid chromatography identified gluconic acid as the main organic acid released by Bacillus sp.SVUNM9.This study isthe first report on the isolation and characterization of native potassium solubilising bacteria from mica ore

Isolation of Amylase Producing Bacteria from Solar Salterns of Nellore District, A ndhra P radesh , India

α-Amylases are a class of starch degrading enzymes catalyzing the hydrolysis of internal α-1,4-O-glycosidic bonds in polysaccharides. The following investigation were carried out to isolate haloalkaliphilic bacteria, a group of organisms with twin extremities of pH and salinity, capable of producing α-amylases from an artificial solar saltern. A total of 25 discrete colonies were isolated, 21 isolates showed amylase production. Among these 7 isolates produced amylase at extreme conditions such as salt, alkalinity and temperature. The isolates were characterized biochemically and also for other enzymes. From the results it is imperative that these isolates can be further studied to exploit them up to industrial scale.

Amylases have potential application in the food fermentation, textile, paper and pharmaceutical industries. There are several processes in the medical and clinical areas that involve the application of amylase. The nutritional and cultural conditions are required for the optimum growth and production of amylase from the mycotic flora. The enzymes are very sensitive to pH and development of an optimal pH control strategy is helpful in obtaining higher protein productivity. The present work deal with the study of effect of pH on fungal amylase production. This comprises isolation and characterization of amylase producing fungi from different soil sources. These fungi were isolated using spread plate technique. The identification of the fungal isolates was done on the basis of morphology and biochemical tests. The probable genera identified were Aspergillus species, Rhizopus speciesand Fusarium species.The enzyme activity was observed for the isolates at pH range of 6.5, 5 and 9 and it was inferred that selection of optimum pH is essential for the maximum enzyme production.It was observed thatAspergillus flavus,Aspergillus nigerand Rhizopus specieswere best amylase producers at different pH range.

Biochemical Characterization o f Phosphate Degrading P seudomonas Cichorii Isolated From Forest Soils In Seshachalam Hills .

Pseudomonas cichoriiisolates wereobtained from forest soils in Seshachalm hills using selective medium. The isolates were screened for phosphate activity and all the isolates were gram negative and showed bright fluorescence under UV light.The cultural and biochemical characteristics confirmed that the isolates were P.cichorii. Carbohydrates utilization profiles confirmed that the isolates were able to utilize Lactose, Xylose, Fructose, Glycerol, Trehalose, Mannitol and Ribose. However, theisolates were unable to utilize glucose and sucrose. The isolates showed positive results for levan production, oxidase and HCN. However they were negative for citrate utilization, ONPG and Gelatin hydrolysis

Ten different Lactobacillus isolates were isolated from marine waters using MRS medium. On MRS broth all the isolates showed uniform growth. All the isolates were Gram +ve, rod shaped bacteria, homofermtative, with creamy smooth round colonies on MRS agar medium. They were Oxidase and Indole negative. Some of the strains are catalase positive. The carbohydrate utilization profile are strain specific. All the isolated strains were unable to utilize Arabinose
