Information on the estimation of genetic distances and differentiation among sheep breeds are needed in crossing and conservation programs. This research aims to study of utilizing behaviour characteristic variables to differentiate and estimate genetic distance between the sheep breeds. The study was conducted at Cilebut and Bogor Animal House of Indonesian Research Institute for Animal Production. Five sheep breeds used were Barbados Black Belly Cross (BC), Garut Composite (KG), Garut Local (LG), Sumatera Composite (KS) and St. Croix Cross (SC), with total sample of 50 heads. A total of 10 variables of behavior traits were observed in this study. Analysis of variances and significance tests were applied to compare between sheep breeds and performed for all of behavior traits using PROC GLM of SAS Program ver. 9.0. PROC CANDISC was used for canonical discriminant analyses, the hierarchical clustering was performed using the PROC CLUSTER by Average Linkage method (Unweighted Pair-Group Method Using Arithmetic Averages, UPGMA), and the dendogram for the five sheep breeds was described using PROC TREE. The differentiator variables for the behavior traits were standing and feeding duration. The canonical plotting based on behavioral characteristics could differentiate BC, KS and LG (with KG and SC) sheeps. Egg production of quails depends on quality of ration. Nutrient manipulation by chromiun inclusion in ration is a possible way to improve production. It is known that chromium mineral in form of GTF in blood has a role not only in enhancement of glucose entering cells through improvement of insulin activity but also in metabolism of lipid and synthesis of protein and elimination of heat stress to improve egg production. This study aimed at assessing egg production of quails fed ration containing chromium-yeast. Sixty-four quails aged 40 days were used. A completely randomized design with 4 treatments and 4 replication was applied in this study. Treatment consisted of commercial ration + Cr 0 ppm (R1), commercial ration + Cr 0.5 ppm (R2), commercial ration + Cr 1 ppm (R3), and commercial ration+ Cr 1.5 ppm (R4). Measurements were taken on feed intake, egg weight, egg mass production, hen day, feed conversion rate, egg index, and egg shell thickness. Results showed that A ration containing organic chromium as much as 1,5 ppm did not affect feed intake, egg production, egg weight, and eggshell thickness, however lowered feed conversion rate by up to 32.25% from that of control. Supplementation of 0,5 ppm chromium in the ration lowered the value of eggs index in the fourth week. Key Words: Quail, Egg Production, Chromium-Yeast

Duckweed (Lemna minor) is a small aquatic plant that grow and float in water and spread extensively. Lemna minor is potential as a source of high quality forage. This study aimed to determine optimal light intensity on Lemna minor to generate maximum productivity. Parameters observed were physical-biological and chemical characteristics of the media (pH value, temperature, cover area, decreased of media volume, BOD, COD, nitrate, nitrite and phosphate), plant growth acceleration (number of shoots, leaf diameter and chlorophyll-a), biomass production, doubling time of cover area and the number of daughters. This study was done based on a completely randomized design with 4 levels of shading. While treatment was: without shading, shading 30%, shading 50% and shading 70% using paranet shade. Each treatment consisted of 4 replications. Result showed that the productivity of Lemna minor included the number of daughters, chlorophyll-a, biomass production, cover area, absorbed phosphate and doubling time the number of daughters reached the highest level without shading treatment (1007,21-2813,57 lux). The decrease of intensity of light, the increase the diameter of leaf. Decrease of media volume was positively correlated to size of cover area. Seroprevalence studies of HPAI H5 subtype in ducks and muscouvy duck in smallholders farm was carried out in Serang and Tangerang District, Banten Province. The study comprised a serological survey to define the distribution and prevalence of HPAI H5 subtype infection on ducks and muscouvy ducks as well as attempted isolation of the virus from these species. Unit of sample in each stage was randomly choosen by multy stage random sample. Blood samples were taken from ducks and muscouvy ducks that had never been vaccinated, purposively. Sera were tested using Haemaglutination Inhibition Test, antigen H5N1 (clade 2.3.2), while the cloaca and trachea swab samples was injected into specific pathogens free (SPF) embryonated 9-11 days old, to isolate the virus. Results showed that H5 subtype virus could be isolated from tracheal swabs of ducks in the various age groups. The seroprevalence of H5 subtype virus in Banten Province was 25.5%, in which, 24.3% occured in ducks and in muscouvy duck in the rate of 1.2%, with titer HI positive was > 3log2. Based on species, seroprevalence level HPAI H5 subtype in ducks was 3-4 times higher than the level of seroprevalence of HPAI H5 subtype in muscouvy duck, which indicated that the H5 subtype virus more likely to circulate in the ducks flock than in muscouvy duck. Diacylglycerol acyltransferase 1 (DGAT1) gene is one of the major genes that has an important role in milk fat synthesis. This research was aimed at to identifying genetic polymorphism of the DGAT1 gene by PCR-RFLP method and its association to milk fatty acid components. Animals studied were Holstein Friesian (HF) cattle from BBPTU Baturraden (123 cows) and BPPT SP Cikole (36 cows). The length of PCR product of the DGAT1gene was 411 bp. Genotyping resulted in two types of alleles, namely K (411 bp) and A (203 and 208 bp); and two genotypes, namely KK (411 bp) and AK (203, 208 and 411 bp). For both locations, genotype frequency of AK (0.75) was higher than KK (0.25). The allele frequency of K (0.64) was higher than A (0.36). Heterozygosity of HF cattles at both locations was relatively high (Ho>He). The DGAT1 gene of the observed HF cattle was polymorphic. Result showed that there was an association between the DGAT1 polymorphism with unsaturated fatty acids especially in nervonat acid. The AK cows had a significant effect on unsaturated fatty acid content of which having a higher nervonat content (0.05%) (P<0.05) than that of the KK cows (0.03%). From the results, it is concluded that the DGAT1 gene can be functioned as a marker of selection for milk fatty acids. Key Words: DGAT1, PCR-RFLP, Holstein Friesian, Fatty Acids

A series of experiment was conducted in order to substitute soybean meal (SBM) with palm kernel cake (PKC) as a protein source in laying hens diet. First experiment was to increase its protein and amino acids content by fermentation process and followed by enzymes supplementation to improve nutrient digestibilities. Second experiment was conducted to evaluate the effectiveness of enzyme- supplemented fermented palm kernel cake (EFPKC) to replace SBM in laying hens diet. The energy (AME) of the PKC, the fermented PKC (FPKC) and the EFPKC was measured by ileal amino acids digestibility (IAAD) in broilers. The AME and the IAAD values of the EFPKC were used for diet formulation in the feeding trial. A feeding trial was performed in laying hens, aged 51 weeks for 8 weeks egg production. Five diets with different levels of substitution of SBM with EFPKC but similar nutrient contents were formulated, i.e.: 1. Control (without EFPKC), 2. 25% of SBM in control diet substituted with EFPKC, 3. 50% of SBM in control diet substituted with EFPKC, 4. 75% of SBM in control diet substituted with EFPKC and 5. 100% of SBM in control diet substituted with EFPKC. Each diet was fed to 24 hens (6 replicates of 4 birds/ replicate). Results of the experiment showed that the fermentation of PKC increased the crude protein and most of the amino acids contents except the threonine and arginine, but decreased its AME. Supplementation of enzymes (BS4 or CE) improved the ileal amino acid digestibilities of the fermented PKC. However, only BS4 enzymes increased the AME of the EFPKC. About 25% to 50% of the SBM in the diet could be substituted with the EFPKC without any detrimental effect on the performances of laying hens. An experiment in feeding pre-starter diets was carried out on local chicken (KUB chicken) raised to the age of 84 days. Four hundred and eighty day-old KUB chicks were allocated to experimental diets of P1 = standard diet without pre-starter; P2 = OASIS® pre-starter for 48 hours feeding; P3 = COBA-1, a mixture of 76.3% yolk powder, 0.76% inulin powder, 7.63 % honey and 15.3% tomato sauce, for 24 hours feeding; P4 = P3 given for 48 hours feeding; P5 = fresh papaya for 24 hours feeding; P6 = P5 for 48 hours feeding; P7 = fasting for 24 hours and P8 = fasting for 48 hours. Following treatment, the chicks were then fed with standard diet, containing 17.5 % crude protein with 2800 kcal ME/kg up to the end of the experiment. Results showed that the group of chicken on pre-starter diet of ripe papaya fruit (P5 and P6), responded better EPEF (European Performance Efficiency Factor) value of 442 and 356 g/bird, respectively in chicken of P5 and P6. Inclusion of crude fiber in diet is important for duck growth, but there is a limit in its use in order for the ducks to grow normally. The purpose of this study was to evaluate growth and carcass production responses of EPMp broiler ducks under different levels of crude fiber and protein in diets. Four hundreds and twenty day old ducklings were allocated into 7 treatments with 6 replications and each replication consisted of 10 ducks. The treatments were the factorial combinations of crude fiber content of 6 or 9% and protein content of 19, 21, or 23%; and BR-1 (starter diet) as positive control. The variables observed were: feed intake, weekly body weight, and percentage of carcass production. The results showed that all variables observed were not significantly affected by CF content, but highly significantly affected by crude protein levels in diet. Protein content of 19 or 21% in diet resulted in a better performance for EPMp ducks. The inclusion of high CF in diet did not affect carcass percentage, except for reduced abdominal fat. The study implies that administration of high CF (6 or 9%) with a protein content of 19 or 21% in the diet are still acceptable to EPMp ducks at 12 weeks. The objective of this study was to develop practical, efficient method for production, purification and assay of binding activity of streptavidin. Streptomyces avidinii was first propagated on agar plates, the bacterial cells on the agar were scrapped and suspended in a defined synthetic media (4.4 ml/cm2). After 7 days agitation on a rotary shaker (200 rpm/min) at room temprature (≈28°C), the bacterial cells in the culture were pelleted. The culture supernatant was concentrated to 1/62 original volume with 75% saturation ammonium sulphate. After intensive dialysis against ammonium carbonate buffer pH 11, the suspension was loaded into an iminobiotin agarose column chromatography. The adsorbed protein (streptavidin) was eluted with sodium acetate buffer, pH 4, and the eluate was concentrated with an ultrafiltration divice and suspended in PBS. The strepatavidin-binding activity was assayed by a competitive ELISA, a competition between streptavidin in the sample and the HRP-streptavidin conjugate for the biotin (biotinyl IgG) immobilised on wells of a microtitre plate. The detection limit of this assay measured 0.16 µg/ml streptavidin. The method developed in this study produced 160 µg/ml streptavidin in the culture supernatant. After concentration with the ammonium sulphate, the streptavidin concentration increased to 4 mg/ml (69% recovery). At the final step of purification, streptavidin with 10 mg/ml concentration was obtained. The purity of the streptavidin was higher (95%) with a recovary of 19%. The purified streptavidin in this study appeared as a dimer core streotavidin on SDS PAGE and its binding activity was twice as high as that of a commercial one. This study was carried out to determine the productivity, competition indices and nutrient content of Brachiaria decumbens (grass), Centrosema pubescens and Clitoria ternatea (legumes) as sole and mixed cropping system in peatland in Pekanbaru, Indonesia from October 2011 to November 2012. The experiment was set up in randomized complete block design with five treatments and three blocks as replication. Five treatments compared: B. decumbens sole cropping (Bd), C. pubescens sole cropping (Cp), C. ternatea sole cropping (Ct), B..decumbens and C. pubescens mixed cropping (Bd+Cp) and B. decumbens and C..ternatea mixed cropping (Bd+Ct). The dry matter (DM) yield of B. decumbens was significantly (P<0.05) increased by mixed cropping. B. decumbens DM yield in C. pubescens intercrop increased by 147.9% and in C. ternatea intercrop increased by 74.1% compare to sole B. decumbens. Land equivalent ratio (LER) value range from 1.04 (Bd+Ct) to 1.58 (Bd+Cp). The crowding coefficient (K) value of B. decumbens in both mixed croping system was higher than K value of C. pubescens and C. ternatea. The total K value for Bd+Cp was higher than Bd+Ct. The competition ratio (CR) value of B. decumbens mixed cropping with C. pubescens and C. ternatea were.>1. The aggressivity (A) value of B. decumbens in both mixed cropping was positive. The crude protein (CP) content of B.decumbens did not significantly (P>0.05) increased by mixed cropping with legumes. Intercropping with B. decumbens significantly (P<0.05) decreased CP content of C. ternatea. Meanwhile neutral detergent fibre (NDF) and acid detergent fibre (ADF) content of B. decumbens did not decrease by intercropping with C.pubescens and C. ternatea. In conclusion, mixed cropping with C. pubescens and C. ternatea in peatland increased DM yield of B. decumbens. Mixed cropping with B. decumbens did not influence DM yield of C. pubescens and decreased DM yield and CP content of C..ternatea. Mixed cropping of B. decumbens with C. pubescens and B. decumbens with C..ternatea in peatland did not increase total DM yield of forage per unit area of land and nutrition contents of forage. Prolactin gene plays crucial role in the reproduction and egg production of birds. The objectives of this study were to characterize single nucleotide polymorphism in partial intron and coding region of duck prolactin gene. Blood samples were collected from 168 ducks consisted of 19 Pekin, 36 Mojosari, and 113 of their crossbreds collected from Indonesian Research Institute for Animal Production (IRIAP). Primer pairs for the coding regions in prolactin gene were self designed based on the duck genomic sequence database (GeneBank: AB158611.1). PCR products based on DNA of prolactin gene exon two was amplified approximately 400 bp. There is one base insertion of Adenin at the position of 2001 bp intron two region of duck prolactin. Homology test based on BLAST method indicated 99% identity with duck refference (Code Access GeneBank: AB158611.1). Adenin composition in all of duck samples was higher than refference. Triplet hydrogen bonds between Guanine and Cytosin pairs was higher than those at duplet hydrogen bonds between Adenine and Thymine. All duck samples were homozigous and monomorphyc. Determination of serum total protein concentration and main fractions (albumin and globulin) can be used as an important diagnostic tool in clinical biochemistry. Several factors can affect the concentration of total protein, albumin, globulin and albumin globulin ratio (A/G). The aim of this study is to obtain serum protein profiles, albumin, globulin and A/G ratio based on breed, age and BCS (body condition score). Blood samples from 160 bulls were collected. Blood chemistry were analyzed by photometer principle using a commercial kit. There were significant (P<0.001) breed variation on total protein, albumin, globulin and albumin globulin ratio. Significant age differences were observed on total protein and albumin concentration (P<0.001), while globulin concentration and A/G ratio were also significant (P<0.05). Amongs groups of BCS, significant difference was verified only in the albumin concentration (P<0.05). The concentration of total proteins, albumins and globulins in the serum of the bulls are higher than standard values for cattle, while A/G ratio is lower. Ant-plant (Myrmecodia pendans) is a unique plant which is widely used in Central Kalimantan and contains antioxidant flavonoids and tannin. The use of herbs as organic materials may produce good performance and carcass of chicken. The experiment used 40 Ross strain day old chicks which were randomly divided into 5 treatments and 4 replicates. The treatments were T0 (basal diet as a control), T1 (basal diet+0.1% ant-plant), T2 (basal diet+0.2% ant-plant), T3 (basal diet+0.3% ant-plant) and T4 (basal diet+0.4% ant-plant). Data were collected during 35 days to obtain feed intake, body weight gain, feed conversion, final body weight, mortality, carcass percentage and performance index. The data were analyzed for variances based on a Completely Randomized Design and continued with Duncan’s Multiple Range Test for differences. The result showed no significant effect of treatments on the performances, but T1 treatment (containing 0.1% ant-plant) showed good tendency to gain body weight, final body weight, carcass percentage and performance index respectively 0.51%; 0.44%; 0.15%; and 3.09% higher than T0 (control). It also showed good feed efficiency with feed conversion of value 2.43% lower than control. A/Duck/Sukoharjo/Bbvw-1428-9/2012 virus subtipe H5N1 clade 2.3.2 as seed vaccine on local duck. AI H5N1 clade 2.3.2 vaccine containing 256 HAU per dose was formulated using adjuvant ISA 71VG Montanide ™. Six groups of one day old local duck were used in this study. Three groups (10 ducks per group) were vaccinated and 3 groups (9 duck per group) were served control. Vaccination was conducted when the duck were three weeks old of age using single dose. Three weeks after vaccination when the duck were challenged either with HPAI H5N1 clade 2.3.2, or HPAI H5N1 clade 2.1.3 virus at dose 106 EID50/ 0.1 ml by drops intranasaly. Result showed that vaccination produced 100% protection compared to unvaccinated ducks againt HPAI subtipe H5N1 clade 2.3.2, and 100% protection againt HPAI H5N1 clade 2.1.3 (A/ck/wj/Subang-29/2007 and A/ck/wj/Smi-Part/2006), while unvaccinated ducks showed virus shedding on day 3 post infection. Morphological characters and agronomic information of Panicum maximum cv Gatton is required in forage crop breeding activities. Therefore a research was conducted to determine the morphology and characteristic of generative phase of Panicum maximum cv Gatton. The study was conducted in greenhouses, using a 40 cm diameter pot. The planting medium was soil from Ciawi (Latosol). Research design was Completely Randomized Design, with 10 replications. Treatments used were 2 types of planting material, 1) derived from the pols (vegetative) and 2 seeds. Plants used were Panicum maximum cv Gatton that have been collected and evaluated at the IRIAP, Ciawi. Variables measured were plant morphology, reproductive traits, seed production per panicle and seed quality. Results showes that morphological characters of cv Gatton derived from seed and pols was not significantly different (p > 0.05), whereas the flowering time and the time of seeds mature after cutting derived from the seeds was faster (p < 0.05) than cultivars derived from pols. Formation of germline chimeras by transfer of chicken primordial germ cells (PGCs) is one of the effective techniques for preservation and regeneration of genetic resources in chickens. This study attempted to form germline chimeras of Gaok chicken buy purifying circulated PGCs of donor embryo before it is transferred to the recipient (White Leghorn chickens=WL) and studied the ability of recipient embryo on survival in incubators, and hatchability. This study used 200 fertile eggs of Gaok and 90 fertile WL breed all of the eggs was incubated at 380C and 60% humidity in a portable incubator. PGCs-circulation of the blood collected Gaok embryos at stage 14-16 were taken from the dorsal aorta, and then purified by centrifugation method using nycodenz. PGCs-circulation results further purification frozen in liquid nitrogen before being transferred to the recipient embryo. The results showed that for the development of embryos transferred to the fresh circulation of PGCs-circulation as many as 25 cells can survive up to day 14, while one of the transferred of 50 and 100 cells into recipient embryos was hatched (10%). On the contrari recipient embryos that are transferred to the frozen PGCs-circulation the embryos development was shorter, and only survived until day 10th (treatment 25 cells), day 14th (treatment of 50 cells) and day 17th (treatment of 100 cells). It is concluded that the amount of PGCs-circulation embryos transferred to the recipient is one factor that influence the success of the development germline chimeras. Milk protein in dairy cattle consists of casein and whey. Whey in milk of dairy cattle is about 20% with the main component is β-lactoglobulin (7-12%). Polymorphism of the β-lactoglobulin gene affects protein and whey production in milk. Selection at a molecular base to improve whey and protein in milk requires information on genetic diversity of the β-lactoglobulin gene, besides other protein genes. This study was aimed to identify genetic polymorphism of the β-lactoglobulin gene in 88 heads of Friesian Holstein (FH) cows at Cikole Dairy Cattle Breeding and Improvement Station (Cikole DCBIS), Lembang. Genotyping was done using Polymerase Chain Reaction - Restriction Fragment Length Polymorphism (PCR-RFLP) method, and a HaeIII restriction enzyme. Genotype frequency, allele frequency, and heterozygosity value were calculated by Nei method. Hardy-Weinberg equilibrium was calculated by chi-square test. Result showed that the β-lactoglobulin gene in the observed HF cattle was polymorphic, in which there were two alleles (A and B) and three genotypes (AA, AB and BB). Frequencies of the A and B alleles were 0.40 and 0.60 respectively; while those of the AA, AB, and BB genotypes were succesively 0.10; 0.60 and 0.30. Heterozygosity value was obtained for 0.483. The β-lactoglobulin gene was in Hardy - Weinberg equilibrium (χ2cal < χ2table). It is concluded that polymorphic of the β-lactoglobulin gene can be used as an initial information for molecular selection on milk protein composition in FH cows. Rabies is zoonotic disease caused by a fatal, neurotropic virus. Rabies virus is classified into the Genus of Lyssavirus under the yang family of Rhabdoviridae. Rabies affecting hot- blooded animals, as well as human. Dogs, cats, monkeys are the vectors or reservoirs for rabies and the virus was transmitted through the saliva after infected animal’s bites. The aim of this study was to conduct rapid diagnosis to detect rabies viral antigen in brain smear using immunohistochemical (IHC) method namely direct Rapid Immunohistochemical Test (dRIT). A total number of 119 brain samples were achieved from Bukittinggi Veterinary Laboratory, West Sumatra. Standardisation and validation of the method were compared to Fluorescent Antibody Test (FAT) as a golden standard for rabies diagnosis. Results show that dRIT was a very good method, it can be performed within two hours without the need of fluorescent microscope. The samples were tested using FAT and from 119 samples tested, 80 (67.23%) samples were positive for rabies and 39 (32.77%) samples were negative for rabies whereas using dRIT showed that 78 (65.54%) samples were positive for rabies and 41 (34.45%) samples were negative for rabies. The dRIT results were validated by comparing them with FAT results as a golden standard for rabies. The relative sensitivity of dRIT to FAT was 97.5% and the relative specificity to FAT was 100% (with Kappa value of 0.976, stated as excellent). The achievement showed that dRIT is very potential diagnostic tool and is highly recommended to be used widely as a rapid diagnosis tool for rabies. B. thuringiensis produces toxic crystal proteine (-endotoksin) which is specific for insect target, but non-toxic to human or non-target organism. Local isolate of B. thuringiensis have been collected from Kediri Regency where endemic area of myiasis. The aim of this study was to formulate the micro-encapsulated of B. thuringiensis using Freund Incomplete Adjuvant (FIA) for protecting crystal in the field and enviromentally friendly. The formulae was then tested in vitro and in vivo. The result of in vitro trial towards seven local isolates of B. thuringiensis showed a decreased in the toxicity of those several isolates. In vivo trial of nine thin tail sheep using chosen isolat of 45.5A and 47.3A, resulted is no significant different for treatment sheep compared to control sheep (P > 0.05). It is concluded that micro-encapsulation method need to be developed according to the way of living the C. bezziana larvae in animal tissues.
