A study has been done to provide a conceptual basis for the development of agricultural system in critical land area in Yogyakarta Special Teritory as the smallest production unit to (1) determine optimum dairy farming based agriculture for the area and income maximization on either farmer own inputs or with CAFlTAL aid provided by a funding party, (2) to develop a model for developing farming system based on dairy farming in critical land area in Yogyakarta Special Teritory.Methods being used multistage problem solving approach to the problem encountered in farming system as practiced in Glagaharjo Village, Sub District of Cangkringan, Sleman with Rapid Rural Appraisal (RRA) and Agro-ecosystem Analysis as diagnostic activities. Farm record keeping and survey to fourty respondents purposively selected were then conducted. Using Linear Programming (LPM), a normative solution, under the imposed constraints for each activity, was obtained to see development effectivity of dairy based farming agricultural system. The result indicated that land resources had not managed optimally. Out of an average possession of 1,5 ha dry land, only 0,6 ha was cultivated, leading to relativelly low yield and income. Optimum dairy farming agriculture migh be an option to develop the area. Dairy farm Cooperative Agency (Model of Scenario I) can play important role to improve farmer income. With an average of 1,5 ha dry land and an average of 3 Animal Unit raised would optimized the resources allocation and income of Rp.20,385,340 for a period of eight years. The income rnigh be further increased to Rp.36,176,070 for similar production period with soft loan provided by ventura fund (Model of Scenario ll) to optimize land resources and family labor force use and a farmer was able to raised an average of 4 Animal Unit. If as being planned - such as scheme is adopted by 1,400 farmer, 230 worker will be employed in the first years. Employement need is increasing with time and after eight year it may reach 1154 workers/year. It can then be concluded that dairy based farming agriculture may improved economic endurance of farmer in critical land area, and directed as well toward land conservation program. Twenty-nine fetuses (11 fetuses from 9 non-superovulated ewes and 18 fetuses from 8 superovulated ewes) were used to study the effect of superovulation of ewes prior to mating on fetal weight, fetal length, the length of the body and limbs, chest circumference, weights of the body, head, neck, limb, and viscera. Superovulated ewes, though with a higher litter size, had a greater fetal growth as was indicated by the greater fetal weight and length, the length and weight of the body and limb on day 49 of pregnancy. On day 105 of pregnancy, superovulated ewes with multiple fetuses (≥3) had similar fetal growth than nonsuperovulated ewes with single and twin fetuses. However, superovulated ewes with a single fetus had greater fetal growth as was shown by the greater fetal weight and length, the length of the body and limbs, chest circumference, and weight of the body, limb, and viscera when compared to those non-superovulated ewes with a single or twin fetuses. Solid substrate fermentation by Aspergillus niger has been carried out to improve the nutritive value of palm oil sludge (POS). POS was fermented aerobically for four days in a fermentor chambers (28°C, RH 80%), with 60% moisture content Some of the product was further incubated anaerobically for 2 days at 28°C. Both products from aerobic and anaerobic fermentation processes were dried by various methods, i.e. sunlight, oven at 60°C, oven with blower at 40°C, at the moisture content less than 11%. Results of the drying methods were also compared with the fresh fermented product. Statistic analysis using factorial design (2 x 4) showed that there was no interaction between kind of fermentation processes (aerobic and anaerobic) and drying methods (fresh, sunlight, oven 60°C, and blower 40°C) for almost all parameters except total a-amino acid content Significant results (p<0.05) were obtained on the drying methods for parameters of crude protein, true protein, in vitro dry matter and protein digestibilities, and mannanase and cellulase activities. There were no significant results between treatments in the crude fiber analysis and soluble nitrogen content Significant results also did not occur between treatment of aerob and anaerob fermentation processes for almost all parameters except for dry matter digestibilities. Results from true protein and in vitro digestibilities show that the fresh fermented product has the best nutritive value, while product dried by sunlight was best among other drying processes. Results from in vivo of protein and energy digestibilities show that there were better metabolizable energy and protein for product with aerobic process and dried with oven and blower treatments, while sunlight drying was best for product processed in anaerobic condition. Although fresh fermented product gave better result from in vitro digestibilities and enzyme activity analyses, for some reasons (easy handling and preservation) sunlight drying gave best results for products processed under anaerobic condition, especially when sunlight drying is cheap. A study on the phenotypic detection of surface antigen of B lymphocytes, MHC I and MHC II in lymph nodes of Bali cattle affected by malignant catarrhal fever (MCF) was conducted by immunohistochemical techniques using avidin-biotin complex peroxidase methods. A number of monoclonal antibodies against surface antigen of B lymphocytes, MHC I and MHC II were used. The results showed that the surface antigens were all detected either in the MCF or in the non-MCF Bali cattle lymph nodes. MHC I were shown predominantly occupied in the lymph nodes of infected cattle. The surface antigen of B cells and MHC II were less found in the infected ones. This indicates that in the immunopathological processes in MCF infected cattle, B cells were not actively involved in producing antibodies, whereas MHC I may contribute the high degree of susceptibility of Bali cattle to MCF. The research was aimed to study factors that affected litter size (JAS), lamb survival (DHA) and litter weight at birth (BL) and at weaning (BS) on prolific ewes. Production records during the year of 1981-1993 were used in this study (1,437 records). Ewe genotypes (G), management levels (M), ewe parity (P) were the fixed effect and ewe gestation gain (PKB) was a covariable that included in the model. Analysis of variance for unbalanced data with general linear model procedure were used. Mean JAS was 1.77. Sources of variation that affected JAS were G, M, P-M interaction, and PKB. One copy of FecF gene increased the number of lambs born by 0.8. Changing toward a better management levels was followed by an increase of JAS. An increased of one kilogram PKB was also followed by an increase of 0.04 litter size born. The mean of BL was 3.43 kg and the sources of variation that affected BL were G, M and P-M interaction. At all management levels, primipara ewes had lower litter size at birth and litter birth weight than the pluripara ewes. Mean DHA found was 73.93%, it was affected by G-M interaction, P and PKB. First parity ewes has 11% lower of DHA than second parity ewes. An increase of 1 kg PKB was followed by an increase of 2.3% of DHA. Sources of variation that affected BS were G-M interaction, P and PKB. Mean BS in this study was 13.12 kg. Key words : Prolific sheep, lamb production

An experiment was done to evaluate the protein-energy requirement for growing Kacang goats. Twelve males and 18 female goats, seven to eight months old were used in this study and randomized into three treatment groups, with four and six animals each, and were kept in individual pens. The treatments used were: R1= Elephant grass (E.G.) + concentrate C1 (21% CP; 3.9 Mcal GE/kg), R2 = E.G. + concentrate C2 (17% CP; 3.7 Mcal GE/kg), and R3 = E.G. + concentrate C3 (12% CP; 3.5 Mcal GE/kg), respectively. Fresh Elephant grass was offered in restricted, and concentrate was offered at 3% of body weight. The experiment was carried out for 12 weeks. Data were analysed by using factorial completely randomized design 2x3 (3 rations and 2 sexes). Parameters measured were: feed intake; average daily gain and feed conversion. The results indicated that among treatments there was no significant difference on dry matter (DM) and gross energy (GE) intake (P>0.05), but crude protein (CP) intake of R1 was 23,6% higher than treatment R2; treatment R2 was 38.1% higher than R3 (P<0.01). Average daily gain (ADG) between treatment R1 and R2, and between R2 and R3 were not significantly different (P>0.05), but treatment R1 was 36.9% and significantly higher than R3 (P<0.01). Feed conversion (FC) between R1 and R2, and between R2 and R3 were not significantly different (P>0.05), but ration R1 was more efficient than R3 (P<0.01). DM intake, GE intake, and ADG of male goats each were 19.4% (P<0.05); 19.0% (P<0.01), and 28.2% (P<0.01) significantly higher than the female goats. Feed conversion between male and female goats was not significantly different (P>0.05). It was concluded that protein intake and average daily gain were increased, and feed conversion was more efficient as the crude protein-energy levels increased in the ration. Feed intake and average daily gain of male goats were higher and feed conversion was more efficient than the female goats. Key words : Protein-energy, Kacang goat

A series of experiment was carried out to evaluate the use of palm kernel meal and its fermented product in the ration of growing ducks. Treatments were arranged in a 2x3 factorial experiment, i.e. the combination between two processes (unfermented and fermented palm kernel meal) and three levels of ingredients in the diet (5, 10 and 15%). Four replicates were applied for each treatment, with 8 ducklings in each replicate. A control diet with no palm kernel meal was also formulated. All diets were formulated to have a similar protein and ME contents. Results of the experiment showed that fermentation process increased the nutrient composition (crude protein, true protein, phosphorus, and the metabolizable energy) and reduced its fibre and fat content. The feeding trial showed that neither the fermentation process, levels of ingredients nor the interactions of the two factors affected final body weight, feed consumption, feed conversion, carcass percentage and abdominal fat of the ducklings significantly. It is concluded that fermentation process improved the nutritional value of palm kernel meal and the inclusion of palm kernel meal (unfermented and fermented), up to 15% in the diet of ducks did not negatively affect the growthperformance and the carcass yield. In order to re-evaluate this evidence, 20 ITT sheep were infected with 350 metacercariae of F. gigantica and for comparison, 10 St. Croix sheep, 10 Merino sheep and 20 crossbred of ITT x St. Croix sheep were also infected with the same dose of metacercariae. The results showed that ITT sheep was highly resistant than the other breed, whereas St. Croix and Merino sheep were susceptible. 60% of the crossbred were as resistant as ITT sheep and the other 40% were as susceptible as the St. Croix sheep. Thus, it is proposed that there might be a hereditary resistance factor such as a dominant gene which inducing the mechanism of resistance in ITT sheep, and there is some indication that IgG2 might act as a blocking antibody that interferes the mechanism of resistance. Myiasis control by means of individual treatment of animals which are mainly rised extensively is time consumed and expensive. The alternative way to control this disease by vaccination is considered effective and economically accepted. However the expected vaccine is now still being developed under a collaborative project between CSIRO, Inter-University Centre on Biotechnology-ITB and Research Institute for Veterinary Science and funded by ACIAR. There are several antigens have been identified as vaccine candidates and an in vitro bioassay technique has been developed for assessing the immunoresponses of vaccine in sheep. Three antigens were used for vaccines in this study, these included protein peritrophic membrane (PM), soluble extract (SE) and pellet extract (PE) of 1st instar larvae of Chrysomya bezziana. Twenty four experimental sheep were divided into 4 groups of 6 animals, 3 groups of animals were injected with PM, SE and PE vaccines with the dose rate of 0.5 g PM/head, 0.8 g PE/head and 4.2 ml LE/head respectively, and the other one group was injected with 4 ml PBS/head as a control group. Vaccination with the same dose was repeated 4 weeks after the 1st vaccination as a booster, and 2 weeks after the booster the sheep were challenged with live larvae, 3 days after challenge animals were killed. Sera were collected at the day of vaccination, 4 weeks after vaccination, 2 weeks after booster, and 3 days after challenge. An in vitro bioassay technique was conducted by culturing 1st instar larvae on five media containing sera collected from each experimental animal. The effects of sera on cultivated larvae were assessed by means of larval weight and larval mortality rate. The results indicated that the growth rate and survival of cultivated larvae in media containing anti-PM sera were significantly lower (P<0.01) compared to the larvae cultivated on media with sera on the day of vaccination. The larval weight depression by anti- PM sera collected at 3 days after challenge was 65% of that larvae cultivated on media with sera collected on the day of vaccination. Anti-PM sera depressed the growth rate and survival of larvae significantly greater (P<0.05) than that of anti-PE or anti-LE sera. The morphological discriminant and canonical analyses were carried out to determine the genetic distance and discriminant variables between Indonesian breeds of thin tail sheep from Deli Serdang (North Sumatera), Ciamis and Garut (West Java) and fat tail sheep from Garahan and Pamekasan (East Java). The number of sheep randomly collected from farmers was 665 heads, while that from breeding station was 183 heads. Zoometrical variables studied were body weight, body length, wither height, chest depth, chest circumference, hip height, hip depth, hip circumference, tail length, tail width and tail thickness. SAS package program was used to analyze the data. Results from distribution mapping produced by canonical analysis and Mahalanobis distance (phenogram tree) showed that breeds are divided into two groups. The first group is thin tail sheep which consists of Ciamis, Garut and Sumatera, and the other is fat tail sheep which consists of Garahan and Pamekasan. The groups of Ciami and Garut were mixed by Garahan with 1.01 and 1.20%, respectively but were not contaminated by Pamekasan (0.00%). The highest similarity between individual inside the group was obtained from Sumatera (84.27%) due to the isolated flock. It was mixed more by Ciamis (7.87%) and Garut (5.62%) than Garahan (1.69%) and Pamekasan (0.50%). Results from canonical analysis also showed that body length, chest circumference, hip circumference, and body weight was less discriminant variables to determine the differences between breeds, while the most discriminant variables were obtained by tail length, tail width and tailthickness. The experiment was conducted to evaluate growth and sexual development of female Etawah-cross kids of the progeny of the does with low (Group L), medium (Group M) and high (Group H) milk production. The does were fed King grass (Pennisetum purpureophoides) (about 2% DM of liveweight) and 400 g/head of concentrate. The amount of concentrate was increased to 600 g/head during the last two months of pregnancy period and during lactation. Kids were weaned at four months of age. Thereafter, they were reared in group and were fed King grass and concentrate. The animals were weighed every two weeks. A mature vasectomized buck were joined in each group to detect the onset of first oestrus (puberty). When kids reached puberty and had a liveweight of about 20 kg, they were mated with intact buck. Ovulation rate, progesterone profiles and fertility were observed. The results showed that pre-weaning growth rates of kids in all three groups were not significantly different (67.0 vs 74.9 vs 70.5 g/day, P>0.05). At six and 12 months of ages, Groups L and M had almost the same liveweight which were higher than Group H. Consequently, Groups L and M reached puberty about 3 - 6 weeks earlier than Group H. The average liveweight at puberty was 19.9, 18.2 and 18.8 kg for Groups L, M and H, respectively, or at about 57.1-69.8% (average 63.2%) of mature liveweight. The pregnancy rate following first mating was relatively high (67-73%) and all does were pregnant in the following mating. Average ovulation rate at conception were low (1.1) in all groups. It was concluded that differences in milk production of the does did not significantly affect post-weaning growth and sexual development and performances of kids. Other factors at certain age or growth phases might be more important to affect growth and reproduction of Etawah-cross goat, and this may warrant further study. feed. Two hundred and ten, day-old broiler chicks were used for this study. They were allotted to 6 different diets containing either BIS or FBIS at 3 different levels (5, 10 and 15%) and one control diet. The results showed that 5% BIS and 5% FBIS could be used in broiler diet without adversely affecting feed intake, weight gain and feed conversion ratio. FCR of those diets were significantly (P<0.05) better than the control diet. Carcass yields were not significantly affected by feeding of BIS nor FBIS. Malignant catarrhal fever (MCF) is a fatal disease affecting large ruminants, some wild animals and pigs. Ovine herpesvirus- 2 (OHV-2) is the causal agent of sheep-associated MCF (SA-MCF). In Indonesia, sheep is epidemiologically implicated in the occurrence of MCF. Providing a sensitive and specific molecular biological technique, i.e. polymerase chain reaction (PCR), a wide range of MCF aspects on molecular level have been developed. The PCR has been successfully used to detect OHV-2 fragment in sheep samples such as peripheral blood leucocytes (PBL) of sheep, nasal secretion of a sheep lamb, and organs of some sheep lambs. Fragment of OHV-2 was also detected in PBL of goats from some areas in Indonesia but not in PBL of pigs from Bali. These results suggest that sheep and sheep lambs are the potential reservoir hosts of MCF in Indonesia, while the role of goats as reservoir host of MCF has yet to be determined. Immunological responses of sheep against adult worm extract antigen of Fasciola gigantica were evaluated in an effort to identify the protein antigen for the candidate of vaccine. In this study the protein antigen from adult worms was extracted, and the extract antigen was then intramuscularly injected into 4 groups of 5 sheep. Two groups received one injection, these included one group injected only with extract antigen and the other group injected with extract antigen emulsified in Quil A adjuvant. The other two groups received two injections with a two week interval, these included one group injected only with extract antigen and the other group injected with extract antigen emulsified in Quil A adjuvant. Three weeks later all of the sheep were challenged with 300 metacercariae of F. gigantica. The antibody titer was monitored every two weeks by using ELISA and the protein profile from each group was compared by using western blotting. Fifteen weeks after challenged all of the sheep were killed and the liver flukes were collected from the liver and counted. The results showed that the antibody titer was higher in the group which received two injections, and the additional of Quil A adjuvant gave much better protection from the infection of F. gigantica (57%) and could avoid the death of the sheep than twice injection of antigen without Quil A adjuvant (37%). Sperm of swamp buffalo bulls is easily damaged during freezing process. Acrosomal intact and plasma membrane intact is important factors in fertilization process. This experiment was aimed to study the effect of freezing method on sperm integrity. The result of experiment indicated that the mean of intact acrosomal and the intact plasma membrane for 4 hours of equilibration (52.24 ± 3.70% and 54.34 ± 4.80%) was significant higher (P<0.05) than 2 hours of equilibration (39.00 ± 3.32% and 43.44 ± 4.91%) but was not significantly difference (P>0.05) with 6 hours of equilibration (47.92 ± 4.51% and 51.58 ± 4.25%). There were not significance difference between one step and two step of glycerolization. A study was conducted to evaluate the responses of lambs on the infection of Haemonchus contortus and the level of energy supplements. Forty eight male lambs, 3-4 months old were used in a factorial experimental design (2x2x2) which lasted for 9 weeks. The factors were breed (Local Sumatra and Sungai Putih: 50% Local Sumatra, 25% St. Croix; 25% Barbados Blackbelly), level of Haemonchus infection (3,000 L3 and 1,500 L3 per week) and level of energy supplement (2,900 kcal/kg DM and 2,300 kcal/kg DM). Infection at 3,000 L3 decreased (P<0.05) ADG of lambs of both energy levels. An extremely low ADG (10-16 g) was observed in 3000 L3 group given low energy level. Body condition of lambs fed high energy level was constant over the experiment, but it decreased at the rate of 0.9 to 1.1 unit at the low energy supplement. EPG was higher (P>0.001) in the 3000 L3 group, was lower (P<0.05) in the Local Sumatra, but not affected (P>0.10) by energy level. PCV was not affected (P>0.10) by infection levels, but lower (P<0.01) in the low energy group or in the St Croix crosses. Eosinophil and serum protein concentration were not affected by energy and infection level, but they were higher (P<0.01) in the local Sumatra. It was concluded that strong effects of level of infection and genotype was detected to influence the impact of Haemonchus infection in lambs. However, the effect of energy levels was less consistent as judged from the EPG, PCV, eosinophil and serum protein data. Escherichia coli isolates showing alpha hemolytic on blood agar were found in calves with bloody diarrhoea from Bandung, Sukabumi and Bogor. Three E. coli isolates designated as (B34c, B909, B910) were pathogenic for mice and selected for vaccine candidates. Crude supernatant of tryptic soy broth (TSB) cultures precipitated with saturated ammoniumsulphate caused cytopathic effect on vero cell line. For vaccine preparation the isolates were grown on tryptic soy agar (TSA). Whole cell inactive vaccine containing equal proportion of each isolate was prepared and emulsified in aluminum hydroxide gel at a final concentration of 1.5% and cell concentration equal to the tube number 10 of MacFarland standard. Mice and rabbits were injected subcutaneously of monovalent vaccine with the dose of 0.2 ml and 1 ml respectively. Four weeks later mice or rabbits were boostered with the same dose. Before injection each animal was bleed, subsequently every two weeks period up to 4 weeks after the second injection. The sera were separated and kept at -20oC up to serological assay with an enzyme linked immunoassay (ELISA) was done. Vaccinated mice with VTEC were challenged with homologous isolate did not die, where as 60% of unvaccinated mice died within 2 days post challenged. Pregnant cows were given 2 doses of three valent vaccine 6 weeks and again 2 weeks before expected date of calving. Calves born were given colostrum and milk of the own mother. The three day old calf was challenged with three isolates of verotoxigenic E. coli studied. Calves born from vaccinated cows did not show diarrhoea at post challenged, whereas calf from unvaccinated cow demonstrated bloody diarrhoea at post challenged. From the experimental animals (mice and calves) demonstrated the presence of protection against challenged. The antiverotoxic antibody probably involved in important role of immunoprotection. These were supported by the presence of high titer anti verotoxic antibody in serum of experimental animals (mice and cows), as well as in experimental rabbits, but none in the unvaccinated animals. The effect of artificial dehydration on snail Lymnaea rubiginosa was investigated in the laboratory by monitoring its survival and development, because as aquatic organism, this snail must be able to adapt or tolerate with the changes in its habitat. Fifty laboratory-reared L. rubiginosa with shell length between 1.0-1.5 cm (adult) were placed in each sloping earth aquarium (60x80x20 cm) which were already filled with water about 15 cm depth. Five aquaria were used in this study, and one week after being established the water from 4 aquaria was drained while one aquarium was retained with water as a control. The water was replaced in one of dehydrated aquarium each week for four weeks, commencing one week after draining the water. The survival of snails in each aquarium were recorded every two days over a period of 3 months. The results indicated that the mortality rate of adult snails increased as the period of dehydration increased. After four week dehydration only 16% of adult snails survive compared to 68% survival in the control aquarium, and dehydration for 4 weeks prolonged the hatching time of eggs. Moreover, in dehydrated aquaria, the egg masses were deposited in a random pattern on the surface of the soil, whereas in the control aquarium they were laid on the soil-water junction. Indonesian climate condition is suitable for growing of mold such as Aspergillus flavus. The mold grown in feed and feed ingredients cause aflatoxins contamination of the feed. Sambiloto has been reported to reduce the growth of Aspergillus flavus and aflatoxins production in feed isolate. The aim of the study was to find the used of sambiloto in reducing aflatoxins contamination in commercial chicken feed. The feed was homogenously mixed then divided into 4 groups with 3 replicates. The samples were treated with sambiloto 0.04%, 0.08% and 0.16% respectively into the feed, except for the control feed sambiloto was not added. Into these feed 4 ml of Aspergillus flavus suspension was also added. Then the feed in each group was incubated at room temperature for 10 days. Samples were collected from each group at days 0, 5 and 10 of incubation times for aflatoxins analysis. Samples were extracted and aflatoxins were detected by High Performance Liquid Chromatography (HPLC). Observation results indicated that sambiloto concentration of 0.16% in feed can inhibit the total aflatoxins production of 16.46% and gave the inhibitation percentage of aflatoxin B1 of 45.39%. Statistical test showed that the used of sambiloto in feed for 5 days incubation times gave significant result in reducing aflatoxins production. However the 10 days incubation times of the feed with sambiloto gave in significant result. It is suggested that to get optimum inhibition of aflatoxins production, sambiloto added in feed should be higher than 0.16%. Key words : Feed, aflatoxin, sambiloto
