Metformin has been known to the mankind for centuries. Metformin is the first line of drug of choice for the treatment of type 2 diabetes. Initial pharmaceutical therapy was for treating the frequent urinal problem which is a primary manifestation of diabetes. At the time of writing, metformin as an intervention is looked into for cardiovascular risks, polycystic ovary syndrome and breast cancer. Annual demand for Metformin is 23,000 Metric tons per annum. Nano Drug Delivery Systems (NDDS) has embarked into Metformin controlled and targeted delivery for breast cancer, pancreatic cancer and non-small cell lung cancer. Clinical estimation of the biological fluids by the FDA approved diagnostic laboratories use LC-MS/MS where blood samples estimate Metformin as an intact molecule easing the pressure of clinical trials result. Metformin was manufactured by a less known and low profile Aoron pharmaceutical, France not aware of the block buster drug hence the potential of technology development start-ups can neither be underwritten nor underestimated even when there is a very high rate (~90%) of failed biotech/biomed start-up ventures. Objective: To ascertain the authenticity of Boeravinone-B in Boerhaavia diffusa and its poly-herbal dosage form (capsule) by identification and quantification through precise and robust HPLC method. Materials and Methods: The RP-HPLC analysis was accomplished employing phosphate buffer & acetonitrile as mobile phase at a flow rate of 1.5 ml/min and 270 nm as detection wavelength. Results: The content of Boeravinone B in ingredients (hydro-alcoholic extracts of whole plant) and poly-herbal formulation of B. diffusa was estimated as 0.041 % w/w and 0.011 % w/w, respectively with good linearity R2> 0.9997 and retention time 24.413± 0.004 min. Conclusion: The amount of Boeravinone- B present in B. diffusa extracts of raw drug complies with that of reported in literature. Quantification of Boeravinone-B suggested an identification mark for this drug and its different market formulations and may work as quality parameter in standardization. On the basis of percentage of Boeravinone-B the content of B. diffusa in poly-herbal formulation validate the ratio of ingredient in final formulation. This method proved its applicability in quality assessment via quantification of this marker in raw material as well as various phyto-formulations of B. diffusa and widens the acceptability criteria at international level.

Objective: The long traditional history of anti-spasmodic effects of Petroselinum Crispum on the gastrointestinal system, and considering histamine receptors in rats ileum, to elucidate the role of the histamine receptor in function relate intestinal motility in this study investigate the effects of histamine and antagonists H2 receptor (Cimetidine) on contractions of adult male rat’s ileum. Methods: In this study, 2 cm of terminal isolated adult male rat’s ileum in an organ bath containing the oxygenated Tyrode solution (370c, PH = 7.4) was used and placed under 1 gram tension contraction induced by 60mM potassium chloride. The effect of parsley extract was measured in presence of histamine and antagonists H2 receptor (cimetidine) and their effects on the rat،s ileum contractions were recorded by using Harvard oscillograph. Results: The parsley extract reduced ileum contraction induced by KCl in a dose-dependent manner (P<0.001, n = 7). Spasmolytic effect of extract after tissue exposure to histamine (0.1 mg/ml) as an agonist and cimetidine (0.004 mg/ml) did not decrease ileum contractions. Conclusion: Although Parsley seeds extract reduced rat ileum contraction, it seems that H2 histamine receptors (cimetidine) are not involved in this activity.
