The seeds of Lactuca sativa L. (Asteraceae) are regarded as an anodyne, galactagogue, demulcent, emollient and used to treat bronchitis, asthma and whooping cough. Phytochemical investigation of the methanolic extract of the seeds of L. sativa led to isolation of phytoconstituents characterized as glyceryl-1,2-dioleiyl-3-phosphate (1), glyceryl-1-octadec-9ʹ-enoate-3-phosphate (2), glyceryl-1-octadec-9ʹ-enoate-2-hexadecanoate-3-phosphate (3), glyceryl-1-octadec-9ʹ-enoate-2-octadecanoate-3-phosphate (4) and β-sitosterol (5). The structures of all the isolated phytoconstituents have been estabilished on the basis of spectral data analysis and chemical reactions.

Floating drug delivery system is one of the novel drug delivery system. Floating drug delivery system has a bulk density less than gastric fluids and thus it remains buoyant in the stomach without affecting gastric emptying rate for a prolonged period of time. Atrovastatin is a competitive inhibitor of HMG-CoA reductase with half-life 14hr. HMG-CoA reductase catalyzes the reduction of 3-hydroxy-3-methylglutaryl-coenzyme A (HMG-CoA) to the mevalonate, which is rate-limiting step in hepatic cholesterol biosynthesis. Inhibition of the enzyme decreses denovo cholesterol synthesis, increasing explanation of low density lipoprotein receptors (LDL receptors) on hepocytes.This increases LDL uptake by the hepatocytes, reducing the amount of low density lipoprotin-cholesterol in the blood. Atrovastatin also reduces blood levels of triglycerides and slightly increases levels of HDL-cholesterol. The floating microsphers of Atrovastatin is being prepared by solvent evaporation method by using ethyl cellulose as a polymer and ethanol as a solvent and after the preparation of microsphere; it’s evaluated for particle size, percentage yield, micromeritic properties, in vitro drug and in-vitro buoyancy release of microsphere, drug polymer compatibility, scaning electron microscopy, incorporation efficiency.

It has been reported that about 40% of the compounds being developed by the pharmaceutical industries are poorly water soluble. The limiting factor to the in vivo performance of poorly water soluble drugs after oral administration their inadequate ability to be wetted by and dissolved into the fluid in the gastrointestinal (GI) tract. Therefore, increasing the dissolution rate of poorly water soluble drugs is an important and significant challenge to pharmaceutical scientists in order to maximize absorption. Aceclofenac is a nonsteroidal anti-inflamatory drug (NSAID), used for rheumatoid arthritis, osteoarthritis and other joint pains. The oral bioavailability of aceclofenac was found to be very poor likely due to the very poor dissolution in aqueous fluids especially in acidic medium. The study utilized the solvent evaporation method for preparation of stable amorphous solid dispersions of Aceclofenac by adsorbing it on porous carrier (Florite).

The purpose of this research was to formulate and optimize an effervescent Gastro retentive tablet formulation of Ondansetron hydrochloride using 32 factorial design. Gastroretentive tablets were prepared by direct compression method by incorporating HPMC K4M, HPC, Carbopol 934P, sodium bicarbonate and citric acid. A 32 Factorial design was applied systemically; the amount of HPMC K4M (X1) and Carbopol 934P (X2) were selected as independent variables. The time required for 50% drug release (T50%), release rate (Ko) and floating lag time (FLT) were selected as dependent variables. It was found that HPMC K4M, Carbopol 934P and their interaction had significant influence on the T50%, release rate (Ko) and floating lag time of the delivery system. The decrease in the release rate was observed with an increase in the concentration of the polymer system. Polymer with lower concentration HPMC K4M was shown to be beneficial than the absence of polymer Carbopol 934P in improving the floating properties of gastroretentive tablet. The observed difference in the drug release and the floating capacity of gastroretentive drug delivery system could be attributed to the difference in the water uptake capacity of the selected polymer.
